Test Principle
The kit is based on the principle of fluorescent PCR technology, and is designed with specific primers and Taqman probes for the detection of RIF/INH resistance mutations in MTB. The single-stranded oligonucleotide sequence that is complementary to the probe sequence is amplified during the PCR process, and the melting curve analysis process is added after the amplification is completed, the melting curve of the hybridization product between the probe and the sequence can be obtained, and the Tm value (melting point) can be obtained, so as to derive the mutation information of the sequence.
The kit includes reaction tubes A, B, C, E, F, and G, wherein the A and E reaction tubes for the detection of isoniazid resistance gene mutations in the KatG gene at codons 315 and 316, and the inhA gene mutations at codons 8 and 15; the B and F reaction tubes for the rpoB gene mutations at positions 511, 513, 516, 531, and 533; the C and G reaction tubes for the rpoB gene mutations at positions 522 and 526.
