Description

1. Principle
Bacitracin ELISA kit is a competitive enzyme immunoassay for the quantitative analysis of bacitracin in the pork, beef and tissue,etc.
The coupling antigens are coated on the micro-well stripes. The Bacitracin  in the sample and the coupling antigens coated on the micro-well stripes compete for the anti-Bacitracin  antibodies. After the addition of the enzyme conjugate, the TMB substrate is added for coloration.
The optical density (OD) value of the sample has a negative correlation with the Bacitracin  in it. The measurement is made photometrically at 450 nm. The absorption is inversely proportional to the Bacitracin concentration in the sample.

Sample preparation

-Tissue, Chicken, pork, porcine liver, shrimp or fish (Lower detect:30ppb)
1.Take 1±0.05 g of the homogenized sample into 10mL centrifuge tube, add 4  mL Redissolving solution, mix thoroughly for 2 min.
2.Centrifuge at above 4000 r/min at room temperature (20-25 ℃) for 5 min.
3.Take 50 µL for analysis.
   Fold of dilution of the sample: 5

-Tissue, Chicken, pork, porcine liver, shrimp or fish (Higher detect:3ppb)
1.Take 3±0.05 g of the homogenized sample into 50 mL centrifuge tube, add 1g NaCI, 6 mL sample Acetonitrile, mix thoroughly with oscillator for 5 min.
2.Take 2 mL supernatant, evaporate to dry at 56℃ in water bath by nitrogen.
3.Add 0.50mL n-Hexane, shake for 30s, add 0.50mL redissolving solution(1×), shake for 30s.
4.Centrifuge at above 4000 r/min at room temperature (20-25 ℃) for 5 min.
5.Remove upper layer, take 50 µL lower layer for analysis.
    Fold of dilution of the sample: 0.50
